The best methods for determining surface roughness in an industrial environment are of the noncontact variety, with reproduction of the profile. The objective of this work was to compare the roughness profile obtained by a contact stylus with a commercial laser displacement sensor (LDS). Measurements were done using 15 wood species with different densities and colors, based on which special triangle profiles were prepared. The accuracy of the laser sensor was examined by statistical analysis of roughness parameters measured from the profiles. Experimental results show that LDS profiles were imitated correctly. However, LDS accuracy depends on the scanned wood properties (density and color), installation position of the sensor, and profile shape. Effective lateral resistance of multiple anchorbolt joints was estimated by considering sill thickness or length/diameter ratios of anchor bolts. Load-slip relationships of single anchor bolt joints were analyzed by the stepwise linear approximation based on the generalized theory of a beam on an elastic foundation and the criterion of “fracture bearing displacement” for several sill thicknesses or length/diameter ratios of anchor bolts. Monte Carlo simulations of the effective lateral resistance of multiple anchor-bolt joints were conducted using the analyzed load-slip curves of single anchor-bolt joints. Effective resistance ratios of multiple anchor-bolt joints were simulated for some combinations of length/diameter ratios of anchor bolts, lead-hole clearances, and number of anchor bolts. The simulated results are: (1) the influence of lead-hole clearance becomes more apparent as length/diameter ratios of single anchor-bolt joints decrease; (2) there is no obvious effect of number of anchor-bolts over the range of 5 to 15; (3) average effective resistance ratios can be adopted for allowable stress design; and (4) reduction of effective resistance ratios should be considered particularly for small length/diameter ratios of anchor-bolt joints.  相似文献

We examined the abrasive wear properties and the effect of abrasive grain size on the rate of wear when sugi wood (Cryptomeria japonica D.Don), compressed to various densities, was rubbed with abrasive paper. The results showed that the wear resistance of compressed wood increased linearly with the increased compression ratio; and under the condition of a low compression ratio it tended to be higher in comparison with the strength of compressed wood. The critical grain size effect, which can be witnessed during the abrasive wear of metals and plastics, was seen when low pressure was applied to the abrasive material. At higher pressures, the wear rate of the compressed wood increased with grain size, but the critical grain size effect was not observed. The pressure required to create the critical grain size effect was found to be higher than that needed for other types of uncompressed wood with the same yield properties.Part of this report was presented at the 50th Annual Meeting of the Japan Wood Research Society, Kyoto, April 2000  相似文献

Cytotoxic T-lymphocyte (CTL) responses to hemagglutinin (H) protein of canine distemper virus (CDV) were evaluated in dogs using the replication-deficient adenovirus protein expression system. Skin fibroblasts were isolated from two dogs and were infected with recombinant adenovirus bearing the CDV-H gene (Ade-CDVH). CTL assay was performed using fibroblasts expressing CDV-H protein as target cells and peripheral blood lymphocytes (PBL) collected from the same dogs one week after immunization of CDV as effector cells. Specific cytotoxic activity was observed against autologous but not heterologous fibroblasts expressing CDV-H protein. These results indicate that the CTL epitope(s) were localized in the H protein.  相似文献

We collected 247 isolates of Cercospora kikuchii from soybean seeds with typical purple stain symptoms from 15 prefectures in Japan. Of the 247 isolates, 93 were sensitive to thiophanate-methyl, a benzimidazole used to control this soybean disease; the remaining 154 were highly resistant to the fungicide. To examine genetic variability among the population of 247 isolates, we developed amplified fragment length polymorphism (AFLP) markers. An AFLP primer pair generated DNA fingerprint polymorphisms among the sample isolates, and with the unweighted pair-grouping method to cluster arithmetic means of the similarity coefficients among all pairs of the fingerprint patterns, the isolates were divided into four lineages (I to IV). Of the 247 isolates, 225 belonged to lineage I, including all isolates that were resistant to thiophanate-methyl. To determine whether the resistance of these isolates was related to mutations in the β-tubulin gene, we amplified partial nucleotide sequences of the gene from 29 representative isolates, including 12 that were resistant to thiophanate-methyl, by means of the polymerase chain reaction. The resistant isolates had identical nucleotide sequence with a one-step change at codon 198, in which the amino acid glutamic acid had been replaced by alanine. The evidence thus suggests that thiophanate-methyl resistance might have arisen in lineage I, the largest of the four lineages. Clearance of Theileria sergenti-infected bovine red blood cells (Bo-RBCs) from the blood circulation of severe combined immune deficiency (SCID) mice was studied to help understand the mechanisms of anemia developing in cattle infected with T. sergenti. For the clearance test, Bo-RBC samples having 2%, 58%, and 76% parasitemia and, as a control, parasite-free Bo-RBCs were prepared in the Bo-RBC-SCID mouse model. The T. sergenti-infected Bo-RBCs and the uninfected control Bo-RBCs were separately labeled with two, green and red, fluorescent dyes, mixed together, and injected intravenously into SCID mice. The blood samples collected at various time points were observed under a fluorescent microscope, and the numbers of green and red fluorescing RBCs were counted differentially to determine the clearance rates of T. sergenti-infected and uninfected Bo-RBCs. This test clearly demonstrated that the Bo-RBC samples having higher parasitemias were cleared faster from the blood circulation of SCID mice. The results suggest that the intravascular clearance system in SCID mice may have a mechanism by which T. sergenti-parasitized and non-parasitized Bo-RBCs are recognized and cleared differentially.  相似文献

ABSTRACT A collection 160 isolates of Cercospora kikuchii was made from South America and 245 from Japan. DNA fingerprint patterns were analyzed based on amplified fragment length polymorphism among the sample isolates, dividing the isolates into seven lineages (I to VII). Partial nucleotide sequence analyses of the beta-tubulin gene supported this division into seven lineages. Lineages I and III commonly existed in South America and Japan. In all, 136 of the 160 isolates from South America and 223 of the 245 isolates from Japan belonged to lineage I, indicating that lineage I was the major lineage in each area; 5 isolates from South America and 8 isolates from Japan belonged to lineage III. Lineages II (12 isolates) and IV (2 isolates) were specific to Japan and lineages V (3 isolates), VI (1 isolate), and VII (15 isolates) specifically existed in South America. These results suggest that the population genetic structure of C. kikuchii was different between South America and Japan, but the dominance of lineage I was common between the two areas.  相似文献
